The development in the Canton district of the city will be delivered by Lovell Construction on behalf of Tirion, a not-for-profit body created jointly by the Welsh Government and Principality Building Society.
Work on creating the infrastructure at the former Wiggins Paper Mill will start shortly and the first homes will be built from this autumn, with the first phase producing 358 homes for sale on the open market and 102 for rent.
The second phase will produce 340 more homes for rental purposes.
Chief executive of the Tirion Group David Ward said: "The fact that both open market and affordable homes are all being built by the same partner means we will achieve a consistent high quality across the whole development with no distinction between the different parts."
Lovell is keen to help set up a national skills centre in the city to ensure some of the 1,000 construction jobs created in the project go to locals via apprenticeships and training schemes.
